Degradation of a synthetic alpha-oligodeoxynucleotide was studied in order to compare its survival with naturally occurring beta-oligodeoxynucleotides in five systems used for antisense hybridization arrest experiments. In contrast to beta-oligodeoxynucleotides, alpha-oligodeoxynucleotides were not detectably degraded over 24 h at 37 degrees C in HeLa cell postmitochondrial cytoplasmic extract or RPMI 1640 with 10% fetal bovine serum, and showed significant survival after 24 h at 37 degrees C in rabbit reticulocyte lysate, fetal bovine serum and human serum.  相似文献

Over the past decade it has become clear that we cannot always explain the observed reduction in leaf expansion rates during drought by measuring the plant's water relations. This has led us to question the possibility of a role for the cell wall and its biochemical machinery in controlling the rate of leaf expansion during drought. However, if we are to reject or modify previous assumptions regarding the control of leaf expansion during drought, then we must offer alternative explanations. This article addresses recent work from this laboratory and in the literature, concerning the involvement of cell wall-enzymes, pH and abscisic acid (ABA) in regulating leaf expansion during water deficit.  相似文献

Connective tissue-activating peptide III (CTAP-III) and neutrophil-activating peptide-2 (NAP-2) are both derived from a common precursor, platelet basic protein (PBP), which is stored in the -granules of platelets and released upon their activation. CTAP-III is an 85-residue peptide which is converted to NAP-2 by enzymic removal of the 15 amino-terminal residues. Both peptides play a role in the early stages of wound healing and inflammation through different activities. We have cloned the cDNA for PBP and expressed constructs coding for the CTAP-III and NAP-2 polypeptides in Escherichia coli. We have purified and renatured these recombinant proteins. The integrity of the recombinant proteins has been ascertained by in vitro bioassays. CTAP-III causes 51% histamine release from the basophilic cell lin KU812 at 10–7 M, whereas NAP-2 only causes 28% release at the same concentration. In assays on human neutrophils, NAP-2 had an EC 50 of 2×10–8 M in chemotaxis, an EC 50 of 3×10–8 M for shape change, and could displace IL-8 from neutrophils with a K d of 7.5×10–9 M. CTAP-III had no activity in these assays. The disulfide bonds have been identified by peptide mapping and sequence analysis, and are in the positions predicted by homology to interleukin-8 and platelet factor 4. Measurement of the molecular mass at physiologic concentrations by gel permeation chromatography has shown that CTAP-III forms predominantly tetramers and dimers, whereas NAP-2 is only dimetric. SDS/PAGE analysis of samples cross-linked with disuccinimidyl suberate support these topologies. We postulate a mechanism for tetramer formation based on the interaction of the amino-terminal extension in CTAP-III involving a helix–helix interaction that could stabilize the association of two CTAP-III dimers.  相似文献

The identity of a patented endophytic bacterium was established by 16S rRNA sequence analysis as a strain of Bacillus mojavensis, a recently erected species within one of the B. subtilis subgroups. This strain of B. mojavensis is antagonistic to the fungus Fusarium moniliforme, an endophytic mycotoxin-producing pathogen of maize and other plants. There are five other species within this subgroup: Bacillus amyloliquefaciens, B. atrophaeus, B. licheniformis, Brevibacterium halotolerans, Paenibacillus lentimorbus, and P. popilliae. The objectives of this research were to screen other isolates of B. mojavensis, B. subtilis, and the other closely related Bacillus species for endophytic colonizing capacity and to determine the in vitro antagonism to F. moniliforme in an effort to survey the distribution of these traits, which are desirable biological control qualities within the Bacillaceae. Antagonism was determined on nutrient agar, and endophytic colonization was established with maize plants following recovery of rifampin-resistant mutants generated from all strains used in the study. The study established that all 13 strains of B. mojavensis, isolated from major deserts of the world, endophytically colonized maize and were antagonists to F. moniliforme. The endophytic colonization of maize by B. subtilis and other species within this subgroup of the Bacillaceae varied, as did antagonism, to F. moniliforme. Thus, this study suggests that endophytic colonization is another characteristic of the species B. mojavensis. The endophytic habit and demonstrated antagonism to the test fungus indicate that isolates of this species might prove to be important biological control organisms where the endophytic habit is desired.  相似文献

Fischer-344 (F-344) rats differ from other common rat strains in that they fail to show any preference for NaCl at any concentration in two- bottle preference tests. Because 100 microM amiloride partially blocks the NaCl-evoked chorda tympani (CT) response in electrophysiological studies, we tested NaCl preference (0.068-0.273 M) in F-344 rats with and without 100 microM amiloride solution as the solvent. A third group was tested with unadulterated NaCl solutions following CT transection. Amiloride had no significant effect on the NaCl preference-aversion function, whereas CT transection significantly reduced NaCl avoidance. These results suggest that the amiloride-sensitive component of the NaCl response is not necessary for F-344 rats to display avoidance of NaCl, but the entire CT input is.   相似文献
